Producción de enzimas microbianas utilizando subproductos de la industria citrícola

La cepa fúngica Acremonium sp. DSM24697 produce la enzima α-ramnosil-ß-glucosidasa (EC3.2.1.168) cuando cultivada en un medio que contiene hesperidina como inductor. Dada la especificidad de sustrato de esta enzima hacia flavonoides 7-O-rutinosilados, y su modo de acción que permite desglicosilar dichos flavonoides en un solo paso, la misma presenta características promisorias para aplicaciones industriales. En este trabajo se utilizaron subproductos de la industria citrícola ('pellets' de naranja y limón) de bajo costo para optimizar la producción de la actividad ***-alfa***ramnosil-ß-glucosidasa. El cultivo sumergido de Acremoniumsp. produjo actividad α-ramnosil-ß-glucosidasa, que se detectó en el sobrenadante de cultivo luego de 5-7 días. Se observó el máximo de producción con las formulaciones hesperidina-pellets en proporción 1:4, alcanzando 15.9 y 8.5 U/L para naranja y limón, respectivamente. El agregado de peptona y extracto de levadura al medio 1:4 hesperidina-pellets de naranja produjo un incremento en la producción del ~77 %. El extracto enzimático obtenido se comparó con glicosidasas comerciales. El análisis zimográfico del sobrenadante de cultivo mostró la presencia de una única banda de actividad con los sustratos 4-metilumbeliferil-rutinósido y hesperidina, correspondiente a la α-ramnosil-ß-glucosidasa. Por otro lado, los cócteles enzimáticos comerciales hidrolizaron xilano, carboximetilcelulosa y todos los sustratos fluorogénicos ensayados, pero no hesperidina. The fungal strain Acremonium sp. DSM24697 produces α-rhamnosyl-ß-glucosidase activity (EC 3.2.1.168) when cultivated in a hesperidin based medium. This enzyme is specific for 7-O-rutinosylated flavonoids, allowing their one-step deglycosylation with promising applications in several industries. The production of α-rhamnosyl-ß-glucosidase activity was optimized using inexpensive by-products of the citrus industry (orange and lemon 'pellets' typically used as cattle feed). The α-rhamnosyl-ß-glucosidase activity was detected in the culture supernatant of Acremonium sp. DSM24697 after 5-7 days cultivation. The maximum production was observed with 1:4hesperidin-pellets ratio, reaching 15.9 and 8.5 U/L for orange and lemon pellets, respectively. The addition of peptone and yeast extract to the 1:4 hesperidin- orange pellets medium increased ~77 % the enzymatic production. The enzyme preparation and bulk glycosidases were compared by zymographic analysis. The commercial enzymes showed hydrolysis of xylan, carboxymethylcellulose and fluorogenic substrates, but they were unable to hydrolyze the flavonoid hesperidin. On the other hand, the supernatant of Acremonium sp. DSM24697 did not show activity against polysaccharides, and presented a single band with the substrates 4-methylumbelliferyl-rutinoside and hesperidin, corresponding to the enzyme α-rhamnosyl-ß-glucosidase.
