Influencia de la densidad celular sobre la producción de componentes de importancia para el establecimiento de la simbiosis rizobio-leguminosa : exopolisacáridos (epss), lipopolisacáridos (lpss), polisacáridos capsulares (kpss) y proteínas celulares totales

Las poblaciones bacterianas regulan coordinadamente la expresión de los genes mediante la producción de moléculas señal difusibles, las que actuarían induciendo la expresión de genes en respuesta a la densidad celular, en un proceso denominado quorum sensing. Este sistema se transforma entonces en un importante componente de los procesos de señalización. En el presente trabajo se estudió si la molécula señal liberada en cultivos rizobianos de diferentes densidades celulares afecta la producción de otros componentes de importancia para el establecimiento de la simbiosis rizobio-leguminosa, tales como exopolisacáridos (EPS), polisacáridos capsulares (KPS), lipopolisacáridos (LPS), y proteínas celulares totales. Todas las determinaciones se realizaron por triplicado para, posteriormente, realizar un análisis estadístico de los datos obtenidos en la determinación de concentración de EPS, KPS y proteínas celulares totales. La determinación de EPS y KPS se realizó por peso seco y por cuantificación de la concentración de hexosas mediante la técnica de Antrona. Para determinar si se presentaban diferencias en las concentraciones de LPS en las diferentes fases comparadas, se realizó la extracción de los mismos mediante la técnica descrita por Hitchock y Brown, seguida de corrida electroforética en geles de poliacrilamida. Por último, la determinación de proteínas celulares totales, se realizó extrayendo las proteínas de las muestras por sonicación y posterior determinación de la concentración mediante la técnica de Bradford. Luego se corrieron las muestras en gel de poliacrilamida con SDS. Los resultados obtenidos en este trabajo, mostraron diferencias en las concentraciones de EPS, KPS y proteínas celulares totales en fase exponencial respecto de fase estacionaria de crecimiento, y también se observaron diferencias en los perfiles electroforéticos de LPS y proteínas celulares totales en las fases comparadas, para los microorganismos utilizados.
