Defectos óseos circulares diafisiarios tratados con matriz ósea desmineralizada en conejos

Los defectos óseos que asientan en las corticales de los huesos largos generan trastornos estructurales pudiendo provocar nuevas fracturas, uniones retardadas, no uniones, osteomielitis y fistulas. La matriz ósea desmineralizada (MOD) posee propiedades osteoinductoras y osteoconductoras capaces de estimular la osteogénesis. El objetivo del proyecto consistirá en evaluar los resultados del tratamiento de defectos óseos circulares experimentales diafisiaros con MOD. Se emplearán 15 (n=15) conejos a los que se les realizará un defecto circular de 3 mm de diámetro en el tercio distal de uno de los fémures. Los defectos de 12 animales (n=12) o grupo tratamiento (GT) serán rellenados con MOD; los otros 3 defectos (n=3) no recibirán tratamiento y constituirán el grupo control (GC). La matriz que se utilizará se obtendrá según protocolo modificado por los autores. Se realizará seguimiento radiológico inmediatamente concluidas las intervenciones y a las 2, 4 y 6 semanas. Al cabo de ese período los conejos serán sacrificados y se procederá a realizar examen histológico de los defectos mediante sistema de puntuación histopatológico de Lane y por luminiscencia fotocrómica, para lo cual semanalmente a los conejos les serán suministrados 30 mg/Kg/IM de oxitetracilina. Los resultados del sistema de puntuación se introducirán en el software Infostat (Universidad Nacional de Córdoba, Argentina) para ser analizada con ANOVA y Test LSD de Fisher Bone defects of the long bone corticals produce structural disorders which can even produce new fractures, delayed unions, non-unions, osteomyelitis and fistulas. Demineralized bone matrix (DBM) has osteoinductive and osteoconductive properties which are capable of stimulating osteogenesis. The aim of this project is to evaluate the treatment of experimental shaft circular defects with DBM. A 3 mm circular bone defect will be carried out in the third distal of the femur of 15 (n=15) rabbits. The defects of 12 animals (n=12) or treatment group (TG) will be filled with DBM; the other 3 defects (n=3) will not receive treatment and will be the control group (CG). The matrix to be used will be obtained from following a protocol modified by the authors. A radiological follow up will be carried out immediately after the surgery and at 2, 4 and 6 weeks. After this period, the bones will be sacrificed and a histological examination of the defects will be performed by means of Lane’s histopathological punctuation system and by photochromic luminescence, for which the rabbits will receive 30 mg/kg/IM of oxitetraciclin weekly. The results of the punctuation system will be analysed with ANOVA and Fisher LSD Test by the Infostat software (National University of Córdoba, Argentina).
